Background

The interleukin-1 (IL-1) signaling pathway is a central driver of innate immunity and inflammation. IL-1 binding to the IL-1 receptor recruits adaptor proteins and the kinase IRAK1, initiating a cascade that activates NF-κB and MAPK signaling and drives transcription of inflammatory genes. One key IL-1-responsive target is the IL-6 gene; its promoter contains an IL-1 response element (IL1RE) and is rapidly and transiently induced by IL-1, TNF-alpha, phorbol esters, and cyclic AMP agonists. Secreted IL-6 in turn activates STAT3 signaling and regulates IL-17 transcription and Th17 cell differentiation, linking IL-1 signaling to chronic inflammation, autoimmune disease, and inflammatory tissue responses.

Product Description & Applications

The IL1RE Reporter Lentivirus is a lentiviral transcription-factor reporter system that provides a sensitive fluorescent or luminescent readout of IL-1 signaling pathway activation in mammalian cells. The construct contains tandem repeats of the IL-1 response element (IL1RE) derived from the human IL-6 promoter, driving reporter expression in response to IL-1 pathway activity. The particles are purified by PEG precipitation and sucrose gradient centrifugation and efficiently transduce difficult-to-transfect cells, including primary and thawed cultures. A constitutively expressed selection marker (blasticidin, hygromycin, puromycin, or zeocin) supports stable reporter cell line generation, and the broad choice of fluorescent and luminescent reporters supports readout by fluorescence microscopy, flow cytometry, or luminometry for studies of inflammatory signaling, cytokine responses, and pathway-modulating compounds.

About This Product

This reporter lentivirus places a BFP2, d2GFP, EGFP, Firefly Luc, Gaussia Luc, GFP, GFP + Firefly Luc, mCherry, Renilla Luc, RFP, RFP + Firefly Luc reporter gene under the control of tandem consensus response elements specific for the IL-1 signaling pathway transcription factor, coupled to a minimal TATA-box promoter and a proprietary upstream enhancer that maximizes signal-to-noise. The constitutively expressed selection marker (Blasticidin, Hygromycin, Puromycin, Zeocin) and/or secondary reporter enables stable polyclonal cell line generation and flexible readout by fluorescence microscopy, flow cytometry, or luminometry.

Stable integration via the lentiviral backbone ensures consistent, clonally representative reporter expression in dividing and post-mitotic target cells — including primary T cells, macrophages, organoids, and cryopreserved material — eliminating the variability inherent to transient transfection. The self-inactivating LTR design and third-generation packaging minimize insertional mutagenesis risk and ensure biosafety classification at BSL-2.
