JH-131e-153 is a diacylglycerol (DAG)-lactone that acts as a small-molecule activator of Munc13-1, targeting its C1 domain, with an activation order of WT>I590≈R592A≈W588A. Because the Munc13-1 C1 domain is homologous in sequence and structure to that of protein kinase C (PKC), JH-131e-153 also activates PKC isoforms, with an activation order of PKCα>Munc13-1>PKCε. It regulates neuronal processes through Munc13-1 and can be used to study neurodegenerative diseases[1]. It has the molecular formula C22H38O5 and a molecular weight of 382.53 g/mol.

Munc13-1 is a target of the diacylglycerol second messenger pathway and contributes to vesicle maturation during exocytosis. It takes part in neurotransmitter release by acting on synaptic vesicle initiation before vesicle fusion, and in the activity-dependent refilling of the easy-release vesicle pool (RRP).
