Compound Overview
KMG-301AM is the acetoxymethyl ester of KMG-301; it accumulates in mitochondria and is then hydrolyzed there to release KMG-301, a fluorescent probe selective for Mg2+ that functions within mitochondria of intact cells. Because the mitochondrial membrane is impermeable to KMG-301, the probe is not released upon depolarization of the mitochondrial membrane potential, and it can report changes in mitochondrial Mg2+ concentration and reveal Mg2+ transport across the mitochondrial membrane during the early phase of a cellular model[1]. It is supplied as a brown to red liquid (C30H28N3O6+, MW 526.56).

In Vitro
Guidelines: the protocol below is a recommended guideline only and should be modified to specific needs[1]. 1. For optical imaging, apply 20 μM KMG-301AM in Hanks' balanced salt solutions (HBSS) to the cells for 10 min on ice, which avoids hydrolysis of the acetoxymethyl ester by esterase in the cytosol. HBSS composition (mM): NaCl 137, KCl 5.4, CaCl2 1.3, MgCl2 0.5, MgSO4 0.4, Na2HPO4 0.3, KH2PO4 0.4, NaHCO3 4.2, D-glucose 5.6, HEPES 5; adjust pH to 7.4 with NaOH. 2. Wash the cells twice with HBSS and incubate for 15 min at 37°C to allow complete hydrolysis of the acetoxymethyl ester form in mitochondria. 3. For simultaneous use of KMG-104AM and KMG-301AM, incubate the cells with 5 μM KMG-104AM in HBSS for 30 min at 37°C, then stain with KMG-301AM. 4. Incubate isolated mitochondria for 20 min at 37°C with 20 μM KMG-301AM in Mitochondria Imaging Buffer (MIB). Wash the mitochondria twice with MIB and incubate for a further 15 min at 37°C. MIB composition (mM): KCl 125, K2HPO4 2, MgCl2 1, HEPES 5, EDTA 0.02; adjust pH to 7.2 with KOH. Alternatively, use Mg2+-free MIB (without MgCl2). 5. Perform fluorescence imaging experiments with a confocal laser scanning microscope system. Excite KMG-301 at 559 nm from a laser diode and observe the signal at 600-700 nm. For simultaneous measurement, excite KMG-104 at 488 nm from an Argon laser and KMG-301 at 559 nm. Separate the fluorescence with a 560 nm dichroic mirror and observe at 500-545 nm and 600-700 nm, respectively.
