In Vitro

L-NIL concentration-dependently inhibits mouse inducible NOS (miNOS) as well as rat brain constitutive NOS (rcNOS), with considerably greater potency toward miNOS. With miNOS and rcNOS, L-NIL gives IC50 values of 3.3 and 92 pM, respectively, which indicates 28-fold selectivity for miNOS. L-NIL is also approximately 6-fold more potent against miNOS than either L-NMA or L-NNA[2].

In Vivo

In mice, renal IR-induced inflammation, oxidative stress and autophagy are prevented by L-NIL (10 and 30 mg/kg, IP)[1].

AdministrationIntraperitoneally at the end of CLP and at 6 h after sepsis induction.
ResultLed to a negligible increase in plasma NGAL compared to sham mice. Led to a significant decrease in both TLR4 and IL1βprotein contents and clusterin transcript. Showed an increase in NFAT5 mRNA levels, as compared with mice treated with vehicle. Promoted a decrease in AR protein expression, as compared with animals treated with vehicle.
