Readout & Interpretation

Lactate dehydrogenase is a stable cytoplasmic enzyme retained by intact cells and released the moment the plasma membrane loses integrity, which makes extracellular LDH one of the most widely accepted surrogates for membrane damage and compound-induced cell death.

Assay Principle

The LDH Cytotoxicity Colorimetric Assay Kit applies a colorimetric format to this measurement, read at OD 460 nm. Hands-on and incubation time is 1 hour.

The Tribo™ LDH Cytotoxicity Colorimetric Assay Kit uses formazan from oxidized WST-8 to measure the activity of LDH released from damaged cells.

This kit is configured for LDH released into culture medium, the standard readout for compound-induced membrane damage.

Q.Which reader is needed?
A.This kit requires a standard absorbance plate reader. The readout is taken at OD 460 nm.
Q.What does this readout tell me about cell health?
A.This kit is configured for LDH released into culture medium, the standard readout for compound-induced membrane damage.
Q.Why measure LDH Cytotoxicity?
A.Lactate dehydrogenase is a stable cytoplasmic enzyme retained by intact cells and released the moment the plasma membrane loses integrity, which makes extracellular LDH one of the most widely accepted surrogates for membrane damage and compound-induced cell death.
Q.Which sample material is validated?
A.The datasheet lists cells, tissue. Other matrices can usually be adapted, but run a spike-recovery check before relying on the values.
Q.How long does the assay take?
A.Combined incubation and handling time is 1 hour. Keep this interval identical across plates in the same study, since timing differences show up directly in the endpoint.
Q.How should the kit be stored on arrival?
A.Store the Reagent at -20°C protected from light. Shelf life: 6 months. Unpack promptly rather than leaving the box at the bench, and return components to storage immediately after each use.
