Disrupting lipid bilayer cell membranes has strong biological effects; for example, pore-forming cytotoxins break membrane permeability, causing release of inflammatory cytokines and tissue damage. Liposomes share the bilayer structure of cell membranes, so they can be used to test compounds that damage membranes.
Kit Components
| Component | BPD100K |
|---|---|
| Liposomal dye | 1.25 ml |
| Color development solution | 1.25 ml |
| Buffer | 7.5 ml |
| DMSO | 1 ml |
Liposomes are made of HSPC : cholesterol : DSPE-PEG2000 (3:1:1 mass ratio), the same lipids as a clinical liposomal drug; the dye is rhodamine conjugated with diethylenetriamine.
Assay Principle
A dye is encapsulated in liposomes, where its fluorescence is quenched. When the bilayer is disrupted, the dye is released and fluoresces at 580 nm (excitation 540 nm) with the color development solution outside the liposome.
