Liposome drug dissolution is the release of an encapsulated drug into the liposome's surrounding medium. In vitro dissolution tests apply chemical and physical stress to simulate this release; for liposomes loaded by pH-gradient remote loading, an ammonium salt is added to speed up release.
Kit Components
| Component | LDD05 |
|---|---|
| Prepacked spin columns | 40 columns |
| 10 x Dissolution buffer | 1.5 ml |
| Elution buffer | 10 ml |
10 x Dissolution buffer: 2 M (NH4)2SO4, pH 5.5. Elution buffer: 10 mM histidine, pH 6.5, 9.2 % sucrose.
Assay Principle
Liposomal drug is incubated in dissolution buffer at a set temperature; at each time point the released drug is removed on a spin column and the drug remaining in intact liposomes is measured by absorbance. Dissolution (%) is calculated against the time-zero sample, and a half-life is obtained by curve fitting.
