Liposomal drug samples can contain free drug that is not inside the liposomes. Drug may leak during storage or after exposure to organic solvent, ultrasound, freezing or heat, and drug loading during production may be incomplete.
Kit Components
| Component | LDE10 |
|---|---|
| Prepacked spin columns | 30 columns |
| Elution buffer | 10 ml |
Enough for 10 liposome samples measured in triplicate (3 columns per sample).
Assay Principle
Spin columns separate intact liposomes from non-encapsulated drug based on differences between liposomes and free drug molecules, including size. The absorbance of the liposome eluate is compared with the total (unseparated) sample to give the encapsulation yield (%).
