The kit tests how stable liposomal drugs stay in plasma or serum from humans or animals. Its ready-to-use spin columns separate intact liposomes from released drug and plasma proteins, so the fraction of drug still held in intact liposomes can be measured.
Kit Components
| Component | SPS20 |
|---|---|
| Prepacked spin columns | 20 columns |
The kit contains only the 20 prepacked spin columns (one column per sample). Plasma or serum, a 0.22 µm low-protein-binding syringe filter, 1.5 ml receiver tubes and the method used to measure the drug in the eluate are not included.
Assay Principle
After incubation with plasma, the sample is spun through a prepacked spin column. More than 95 % of plasma proteins, together with free and protein-bound drug, stay on the column, while intact liposomal drug passes into the eluate. Recovery of intact liposome is the drug concentration in the eluate of the plasma-treated sample divided by that of a buffer control kept on ice. In the manufacturer's example, liposomal doxorubicin gave 96 % recovery after 2 hours in human plasma at 37 °C.
