Safety

For Research Use Only. Not for use in diagnostic or therapeutic procedures, and not for human or veterinary use. Handle in accordance with the Safety Data Sheet and your institution's chemical hygiene plan.

In Vitro

SolventSolubilityNotes
DMSO100 mg/mL (329.72 mM)requires sonication and warming and heat to 60°C; use freshly opened DMSO (absorbed moisture lowers solubility)

Aliquot the stock solution and store it at -80°C (up to 6 months) or -20°C (up to 1 month); protect from light; avoid repeated freeze-thaw cycles.

In Vivo

Choose the formulation that suits the animal model and route of administration; percentages are volume ratios of the final working solution. Start from a clear DMSO stock (see In Vitro above), add the co-solvents one at a time in the order listed, mixing after each addition, and prepare the working solution fresh on the day of dosing. If precipitation or phase separation occurs, gentle warming or sonication can help.

Protocol 1

Composition2% DMSO + 40% PEG300 + 5% Tween-80 + 53% saline
Result0.5 mg/mL (1.65 mM); suspension; requires sonication

Data provided by the manufacturer.

In Vitro

LLL12 (0.01-1 μM; 72 h) lowers the viability of ovarian cancer cell lines, in the presence or absence of Cisplatin (0.5 μM, 2.5 μM) together with Paclitaxel (0.25 μM, 0.5 μM)[1]. LLL12 (0.25-1.0 μM; 72 h) suppresses STAT3 phosphorylation (Tyr705) in ovarian cancer cell lines[1].

Western Blot Analysis[1]

Cell LineA2780, SKOV3, CAOV-3 and OVCAR5 ovarian cancer cell lines
Concentration0.25, 0.5, and 1 μM for A2780 and OVCAR5; 1, 2.5, and 5 μM for SKOV3 and CAOV-3
Incubation Time72 hours
ResultInhibited STAT3 phosphorylation at Tyr705.

In Vivo

LLL12 (5 mg/kg; i.p.; once daily for 13 d) exerts strong growth-inhibitory activity against mouse osteosarcoma cells and tumor growth[2].

Animal ModelMouse xenografts with SJSA or OS-33 osteosarcoma cells[2]
Dosage5 mg/kg
AdministrationIntraperitoneal injection; once daily for 13 days
ResultResulted in a significant reduction in tumor volume and tumor mass in the OS-33 and SJSA xenografted mice.
