Product Overview
This kit is designed for purifying and enriching DNA and RNA from pathogenic microorganisms, such as viruses, bacteria, and fungi, present in biological fluid samples like sputum, bronchoalveolar lavage fluid, blood, and ascites. The purified microbial DNA and RNA are suitable for a variety of downstream applications, including PCR, RT-PCR, qPCR, NGS, and other molecular biology experiments.
The protocol mixes 400 µL of sample with 200 µL of Buffer LBS-A and 20 µL of Proteinase K, vortexed for 10 min or thermomixed at 65°C and 2500 rpm for 10 min. A second digestion with 250 µL of Buffer ML and 20 µL of Proteinase K runs at 56°C for 10 min, followed by clarification at 12,000 rpm for 1 min. Nucleic acids are bound with 400 µL of isopropanol and 30 µL of Magbeads PN at 1500 rpm for 3 min, washed twice with 800 µL of Buffer GW1 and twice with 800 µL of Buffer GW2, air-dried about 3 min and eluted in 70 µL of RNase-Free Water. For cultured samples the capacity is 1×10⁹ bacterial or fungal cells or fewer.

Storage & Handling
All components are stored at room temperature (10–30°C) with a shelf life of 1 year.
Usage Notes
- Inspect the buffers for precipitate and dissolve them at 56°C if needed.
- Store samples at 2–8°C for the short term or at -20°C or -80°C for the long term.
- Avoid repeated freeze-thaw cycles of the sample.
- Maintain a clean work area and use DNA/RNA-free tips.
- Implement contamination controls throughout the procedure.
- Isopropanol is required at the binding step and is not supplied.
- Store the eluted nucleic acid at -20°C.
