Product Overview

This kit provides a simple and efficient plant DNA extraction solution. DNA in the sample binds to the surface of silica-coated magnetic beads in the presence of high salt. After rinsing, the DNA is eluted in Buffer TB or deionized water. The extraction rate is closely related to the sample type and the cell disruption effect. The extracted DNA can be used for next-generation sequencing, PCR test and other downstream experiments.

The protocol processes 50–100 mg of fresh material or about 30 mg of dry material. The sample is lysed in 400 µL of Buffer PLS with 5 µL of RNase A at 70°C and 1600 rpm for 10 min, clarified at 12,000 rpm for 4 min, then bound with 550 µL of Buffer ZB and 20 µL of Magbeads for 5 min. The beads are washed twice with 650 µL of Buffer GW1 and twice with 650 µL of 75% ethanol, air-dried for 5–10 minutes and eluted in 100 µL of Buffer TB at 65°C for 10 min.

Usage Notes

  • Magbeads PN must not be frozen or subjected to high-speed centrifugation.
  • For plants rich in polysaccharides and polyphenols, β-mercaptoethanol is added to 5%; it is not supplied with the kit. See the manufacturer's manual for the buffer to which it is added.
  • Prepare the 75% ethanol wash separately, as it is not supplied.
  • The extraction rate depends on the sample type and the cell disruption effect.
  • Store the eluted DNA at -20°C.
