In Vitro
Guidelines[1] (the protocol below is a recommended guideline only and can be adapted to specific needs). Labeling of Cells: 1. Incubate the cells following the standard protocol. 2. Pre-incubate the cells with 20 μM MAPTAM for 30 minutes. 3. Exchange the medium for HEPES buffer supplemented with 1% BSA (wt:vol) and 2 μM fura 2 pentaacetoxymethyl ester. 4. Carry out measurements in HEPES buffer containing 1 mM CaCl2.
