MDBN is an irreversible inhibitor of the AAA-ATPase p97, also known as valosin-containing protein (VCP), with an IC50 below 10 µM. p97 plays a central role in protein homeostasis, including ER-associated degradation (ERAD), autophagy, and stress granule dynamics. Mutations in p97 are linked to neurodegenerative disorders such as inclusion body myopathy and frontotemporal dementia. MDBN offers a pharmacological approach to modulate p97 activity and investigate its role in neuronal proteostasis and neurodegeneration.
