Metacetamol, also known as AMAP, is an analog of Acetaminophen. It induces dose-dependent necrosis in primary hepatocytes through glutathione depletion, mitochondrial damage, and formation of mitochondrial protein adducts. Its derivatives act as anticancer and antibacterial agents, and it can be used in studies of breast cancer, bacterial infections, and fungal infections (candidiasis)[1][2]. In Vitro
In primary hepatocytes, Metacetamol (AMAP) (5-20 mM; 24-48 h) causes dose-dependent necrotic cell death and glutathione depletion, and after 48 h of incubation significant cytotoxicity is seen at 5 mM, 10 mM, and 20 mM[1]. At 5-20 mM, Metacetamol (AMAP) also causes dose-dependent mitochondrial dysfunction in primary hepatocytes[1]. Exposure of primary hepatocytes to Metacetamol (AMAP) (6-24 h) leads to significant formation of mitochondrial protein adducts[1].
