Methylglyoxal (MG) is a highly reactive alpha-oxoaldehyde generated as a byproduct of glycolysis and lipid peroxidation. In the nervous system, MG contributes to oxidative stress by forming advanced glycation end products (AGEs) through reactions with amino acids and protein residues. These AGEs disrupt protein function, promote inflammation, and impair cellular signaling—hallmarks of neurodegenerative diseases.
MG also crosslinks DNA polymerase and substrate DNA, inhibiting DNA replication and repair. This genotoxicity exacerbates neuronal damage and accelerates disease progression in conditions such as Alzheimer’s and Parkinson’s disease. Elevated MG levels have been observed in aging brains and in models of metabolic dysfunction, linking it to both neurodegeneration and systemic disease.
Therapeutic strategies aimed at reducing MG accumulation or enhancing its detoxification via the glyoxalase system are under investigation. As a biomarker and mediator of oxidative damage, MG represents a critical target in the study and treatment of neurodegenerative disorders.
A 1:1000 dilution of SMC-516 was sufficient for detection of Methylglyoxal in 0.5 µg of Methylglyoxal conjugated to BSA by ECL immunoblot analysis using Goat Anti-Mouse IgG:HRP as the secondary Antibody.
