mGluR5 (metabotropic glutamate receptor 5) is a subtype of glutamate receptor that regulates synaptic transmission and plasticity via G-protein-coupled signaling. It is predominantly expressed in the hippocampus, cortex, and striatum—regions vulnerable to neurodegeneration.
Aberrant mGluR5 activity has been linked to excitotoxicity, calcium dysregulation, and neuroinflammation, contributing to the pathophysiology of Alzheimer’s disease, Parkinson’s disease, and amyotrophic lateral sclerosis (ALS). mGluR5 also interacts with amyloid precursor protein and tau, influencing their aggregation and toxicity.
Pharmacological modulation of mGluR5 has shown promise in preclinical models, where antagonists reduce neuronal loss and improve cognitive outcomes. As a key regulator of glutamatergic tone, mGluR5 is a compelling target for therapeutic intervention in neurodegenerative disorders characterized by synaptic dysfunction and excitatory imbalance.
1 µg/ml of SMC-419 was sufficient for detection of mGluR1/5 glutamate receptor in 20 µg of rat brain membrane lysate and assayed by colorimetric immunoblot analysis using goat anti-mouse IgG:HRP as the secondary antibody.
