Product Overview
The Micro Phosphate Removal Column Set removes inorganic phosphate from buffer solutions. Phosphate binds to the column resin and stays on the column, while regular buffer salts and non-charged molecules such as sugars and glycerol do not bind and stay in the sample.
Some organic phosphates such as ATP also bind to the resin, but less strongly than inorganic phosphate. Metal chelators such as EDTA interfere with phosphate binding to the resin. Removing phosphate is useful, for example, before phosphatase activity assays.

Protocol Outline
- Spin each column briefly to settle the resin, remove the bottom tip and cap, and spin twice at 13,000 rpm for 1 min so the resin is almost dry.
- Move the column to a clean 1.5 ml tube, load 200 µl of sample, spin at 1,000 rpm for 1 min and then at 13,000 rpm for 1 min, and keep the eluate.
- Spin the eluate at 13,000 rpm for 1 min, or filter it through a 0.22 µm filter, to remove any insoluble material.
The full ProFoldin protocol “Micro Phosphate Removal Column Set” is linked under Documents.
Notes
- Avoid EDTA and other metal chelators in the sample; they interfere with phosphate binding to the resin.
