Enzyme Activity Assay Kit

Kinases transfer a phosphate from ATP to a substrate, giving a phosphorylated substrate and ADP. Measuring the ADP formed is a general way to follow the activity of purified kinases regardless of the substrate.

Pack Sizes

Both sizes contain the same reagents. The only difference is the number of assays: MUK100K is for 100 assays and MUK1000K for 1000 assays (384-well format); see the Kit Components table for the volumes of each size.

100 assays (MUK100K) 100 assays; enzyme not included
1000 assays (MUK1000K) 1000 assays; enzyme not included

Kit Components

Component 100 assays (MUK100K) 1000 assays (MUK1000K)
10 x Assay buffer 600 µl 4000 µl
100 x ATP 35 µl 350 µl
100 x MUK Reagent A 35 µl 350 µl
100 x MUK Reagent B 35 µl 350 µl
1 mM ADP 30 µl 200 µl
10 x Fluorescence dye 350 µl 3500 µl

The 1 x MUK Buffer is 50 mM Tris-HCl, pH 8.0, 50 mM NaCl, 3 mM MgCl2, 0.5 mM DTT, 0.2 mM EDTA, 0.003% Brij-35. Kinase and kinase substrate are supplied by the user.

Assay Principle

The kit measures the ADP produced by the kinase reaction. MUK Reagents A and B are added after a 2 min pre-incubation and are present during the 60 min reaction; a fluorescence dye is then added to read the ADP signal at 535 nm (excitation 485 nm). Low ATP (20 µM recommended) keeps the ADP background low and lets ATP-competitive inhibitors be detected.

Safety & Handling

For Research Use Only (RUO). Not for use in diagnostic or therapeutic procedures. The MicroMolar Universal Kinase Assay Kit is shipped frozen. Follow standard laboratory safety practices when handling the reagents. Please contact us to confirm storage conditions.
Q.What does this kit measure?
A.It measures the activity of purified kinases by detecting the ADP they produce, in the 0.1 µM to 10 µM range. It works with purified ADP-producing kinases and substrates supplied by the user and can be used to test ATP-competitive inhibitors.
Q.What is the difference between the options?
A.Both sizes contain the same reagents. The only difference is the number of assays: MUK100K is for 100 assays and MUK1000K for 1000 assays (384-well format); see the Kit Components table for the volumes of each size.
Q.Is a protocol available?
A.Yes. The manufacturer's protocol (Universal Kinase Assay, linked under Documents) covers catalog # MUK100K, MUK1000K.
Q.How long does the assay take?
A.About 62 min (2 min pre-incubation + 60 min reaction). See the protocol for the exact steps.
Q.How should the kit be stored?
A.The MicroMolar Universal Kinase Assay Kit is shipped frozen. Please contact us for long-term storage conditions.
