(-)-Eseroline fumarate, also known as Eseroline fumarate, is a metabolite of physostigmine, an AChE inhibitor. It causes leakage of lactic acid dehydrogenase (LDH) from cancer cells and induces the release of adenine nucleotides and 5-hydroxytryptamine (5-HT) from neuronal cells, thereby inducing cell death. It also inhibits the electrically evoked twitches of the mouse vas deferens and of the guinea-pig ileum[1][2]. It is supplied as a white to off-white solid (C17H22N2O5, MW 334.37) at 98.03% purity.

In Vitro
(-)-Eseroline fumarate (0.5 mM; 0-25 hr) causes time-dependent LDH leakage from neuronal cell culture systems, namely mouse neuroblastoma N1E-115, as well as rat glioma C6 and neuroblastoma-glioma hybrid NG 108-15[1].
In Vivo
In adult mongrel cats, a single dose of (-)-Eseroline fumarate (HCl; 10 mg/kg; i.v.) induces release of 5-HT from the cerebral cortex[2]. A single dose of (-)-Eseroline fumarate (salicylate; 9 mg/kg; s.c.) has an analgesic effect and strongly reduces the antinociceptive effect of Physostigmine in the mouse tail-flick test[3].
