Product Overview
The miRNA extraction kit is specially used to separate and purify miRNA from various animal tissues, plant tissues, cells, serum, plasma and other samples. It can also extract siRNA, snRNA and other small molecular RNAs less than 200 nt as well as total RNA. This product combines phenol/guanidine lysis technology and silica matrix membrane purification technology. Its unique lysis solution can effectively inhibit RNases while removing most of the DNA and protein in cells or tissue samples by organic extraction. This product can be directly used for Northern Blot analysis, Real-Time PCR, Microarray Analysis, etc.
Two protocols are supplied, one for miRNA enrichment and one for total RNA extraction. The general workflow lyses tissue or cells with TRIzon Reagent, extracts with chloroform, precipitates with ethanol, binds and washes on the supplied spin columns and elutes in 30–50 µL of RNase-Free Water. All centrifugation is carried out at 12,000 rpm at room temperature, and a drying step precedes elution to remove residual ethanol that would otherwise affect enzymatic reactions.

Applications
- miRNA separation and purification from tissues, cells, serum and plasma
- Extraction of siRNA, snRNA and other small RNAs less than 200 nt
- Total RNA extraction
- Northern Blot analysis and Real-Time PCR
- Microarray analysis
Storage & Handling
TRIzon Reagent is stored at 2–8°C in the dark; the other components are stored at room temperature (15–30°C). The shelf life is 1 year at room temperature.
Usage Notes
- Use RNase-free plastics, autoclaved glassware and RNase-free water to prevent RNase contamination.
- Wear a mask and gloves during the procedure.
- Avoid repeated freeze-thaw cycles.
- Add ethanol to the concentrated buffers before first use.
- Do not omit the drying step, as residual ethanol affects enzymatic reactions.
- Chloroform and ethanol are required but are not supplied.
