Biological Activity
MMP-9 inhibitor I is an inhibitor of matrix metalloproteinase-9 (MMP-9) that is selective over MMP-1 and MMP-13 (IC50s = 5, 1,050, and 113 nM, respectively). It also decreases the activity of TNF-alpha converting enzyme (TACE) in a dose-dependent manner (IC50 = 0.54 uM). MMP-9 inhibitor I decreases TNF-alpha secretion stimulated by LPS in BV-2 microglial cells when used at concentrations of 50 and 100 uM.

Use a bath-type ultrasonic cleaner rather than a high-intensity ultrasonic disruptor. Bath sonicators provide gentle, uniform agitation appropriate for dissolving solid compounds without generating cavitation damage or excessive heat. Ensure the solvent tube is fully submerged and use short sonication pulses of 5–10 seconds.
Try the following steps: (1) warm the DMSO solvent to 37–50 °C before adding the compound; (2) vortex vigorously for 30 seconds then bath-sonicate for 1–2 minutes; (3) if still undissolved, prepare a more dilute stock.
