Complement component C3a anaphylatoxin is a multifunctional proinflammatory mediator produced by complement cascade activation and causes inflammatory responses and cytokine production in septic shock, allergy, trauma, ischemic heart disease, pain, neurodegenerative diseases, and several other autoimmune diseases. C3a is rapidly cleaved to the C3a-DesArg form by the endogenous serum carboxypeptidase enzyme.
Assay Principle
The C3a ELISA Kit is for the in vitro determination of mouse C3a in different tissue samples including skin, muscle, nervous system and other biological samples. The microplate is coated with murine C3a monoclonal antibody. C3a present in the sample or standard binds to the antibodies absorbed by the microwells. A biotin-conjugated anti-murine C3a antibody is added and binds to murine C3a captured by the first antibody. Next, streptavidin-horseradish peroxidase (HRP) is added, producing an antibody-antigen-antibody “sandwich”. Finally, substrate solution reactive with HRP is added to the well and exhibits a blue color that is directly proportional to the amount of C3a in the initial sample. The reaction is terminated by the addition of an acid, and absorbance is measured at 450nm. The concentration of C3a in the samples is determined by comparing the O.D. of the samples to the standard curve.
Mouse C3a ELISA Kit is a Solid-phase sandwich ELISA for the quantitative determination of C3a. The microplate is pre-coated with a capture antibody specific to the target. Target present in the sample or standard binds the immobilized antibody, is detected with a biotinylated detection antibody and streptavidin-HRP, and quantified colorimetrically against a standard curve.
