Assay Principle
This sandwich ELISA quantifies mouse insulin-like growth factor-binding protein 1 (gene Igfbp1) in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.
IGFBP-1 is a secreted insulin-like growth factor binding protein produced mainly by the liver and by decidualized endometrium. It binds IGF-1 and IGF-2 and controls their access to receptors, with levels acutely regulated by insulin and nutritional state, and it can also signal through integrin α5β1 to promote cell migration.
