What is the purpose of an isotype control in Flow Cytometry?

An isotype control establishes the background fluorescence threshold from non-specific antibody binding (Fc receptor engagement, hydrophobic interactions) — not antigen-specific binding. Clone MOPC-21 carries no antigen specificity, so all signal it produces represents background noise rather than target detection.

How do I use this isotype control in my flow cytometry panel?

Run MOPC-21 IgG1k PE/TR at 5 µL/Test — the same concentration as your specific PE/TR-conjugated mouse IgG1k antibody of interest — on the same cell population with the same staining protocol. Set your positive gate just above the upper boundary of the MOPC-21 fluorescence distribution.

Why must I not freeze this antibody?

This antibody is conjugated to a PE/TR tandem fluorochrome. Freezing disrupts the energy transfer between PE (donor) and Texas Red (acceptor) chromophores, resulting in loss of far-red (Texas Red channel) emission and appearance of spectral artifacts in the PE channel. Store at 2–8°C only.

What sizes are available?

Which laser and filter should I use to detect PE/TR?

PE/TR (Phycoerythrin/Texas Red tandem) is excited by the 488 nm blue laser. Emission peaks in the Texas Red channel at ~613–615 nm. Use a 610/20 or 613/20 bandpass filter for optimal detection on a standard flow cytometer equipped with a 488 nm laser.
