Scientific background
MAPK15 (Mitogen Activated Protein Kinase 15) is associated with intracellular signaling regulation, often within phosphorylation-dependent pathways that control activation, proliferation, or differentiation.
While kinase activity is frequently assessed by phospho-readouts, total protein abundance can also shift with pathway rewiring, feedback, or changes in cell composition.
Protein quantification can complement phospho-specific assays and help interpret whether signaling changes are driven by abundance, activation state, or both.

How the ELISA works
Designed for Mouse samples, this kit uses a The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Mouse MAPK15. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Mouse MAPK15. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Mouse MAPK15, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Mouse MAPK15 in the samples is then determined by comparing the OD of the samples to the standard curve.. After binding and washing, signal is converted to concentration using a standard curve.
