Scientific Background
Polyamine Oxidaseis an enzyme belongs to the family of oxidoreductases, specifically those acting on the CH-NH group of donors with oxygen as acceptor. Widely expressed. Not detected in spleen. Expressed at lower level in neoplastic tissues. Flavoenzyme which catalyzes the oxidation of N(1)-acetylspermine to spermidine and is thus involved in the polyamine back-conversion. Can also oxidize N(1)-acetylspermidine to putrescine. Substrate specificity: N(1)-acetylspermine = N(1)-acetylspermidine > N1,N(12)-diacylspermine >> spermine. Does not oxidize spermidine. Plays an important role in the regu.
Assay Principle
This assay employs a two-site sandwich ELISA to quantitate PAOX in samples. An antibody specific for PAOX has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPAOX present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PAOX is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PAOX bound in the initial step. The color development is stopped and the intensity of the color is measured.
