MRL-494 is an antibacterial agent that inhibits β-barrel assembly machine A (BamA) and is unaffected by efflux and the outer membrane permeability barrier. It can inhibit Gram-positive bacteria, with an MIC of 12.5 μM for Staphylococcus aureus COL, and Gram-negative bacteria, with an MIC of 25 μM for E. coli JCM158[1]. In Vitro

MRL-494 disrupts the cytoplasmic membrane in a lethal manner. By targeting BamA, MRL-494 inhibits the biogenesis of OM proteins (OMPs) from outside the outer membrane (OM). MRL-494 has strong anti-microbial activity against both Gram-positive and Gram-negative bacteria. Its MIC values are 25 μM against E. coli (WT), 25 μM against E. coli (ΔtolC), 25 μM against E. coli (ΔtolC envA101), 100 μM against K. pneumonia, 200 μM against A. baumannii (WT), 200 μM against A. baumannii (ΔlpxC), 100 μM against P. aeruginosa (efflux deficient), 100 μM against P. aeruginosa (WT), 12.5 μM against Staphylococcus aureus (methicillin-resistant), and 25 μM against Bacillus subtilis rpoB18[1].
