mRNA Cap 2´-O-Methyltransferase is a recombinant protein derived from vaccinia virus. The enzyme can add a methyl group at the 2´-O position of the first nucleotide of the cap structure at the 5´ end of the RNA. The enzyme uses SAM as a methyl donor to methylate capped RNA to form a Cap1 structure.

mRNA Cap 2´-O-Methyltransferase is a recombinant protein derived from vaccinia virus. The enzyme can add a methyl group at the 2´-O position of the first nucleotide of the cap structure at the 5´ end of the RNA. The enzyme uses SAM as a methyl donor to methylate capped RNA to form a Cap1 structure. The Cap1 structure can enhance the translation efficiency of mRNA, so it can improve the expression of mRNA in transfection and microinjection experiments. This enzyme requires RNA with m7GpppN, a 7-methylguanosine cap structure, as a substrate.
This product is produced in accordance with GMP process requirements. The product is provided in liquid form and can be used for Cap1 capping reaction of pre-mRNA in vivo/in vitro.

Store this enzyme at -20°C and avoid repeated freeze-thaw cycles to preserve catalytic activity. The product is shipped Dry Ice and remains stable for up to one year from the date of manufacture when stored under recommended conditions. Aliquoting the stock solution into single-use volumes is recommended for enzymes used infrequently to minimize thermal cycling of the bulk stock.

This enzyme is validated for use in In Vitro Transcription applications. It is suitable for use in both standard laboratory research settings and, where applicable, optimized reaction workflows requiring high specificity, sensitivity, or throughput.

Capping efficiency (%) is determined by the proportion of RNA transcripts bearing a 5′ cap structure following the standard capping reaction. Activity is also expressed as units (U) per µL under manufacturer-defined assay conditions.

This enzyme is produced as Recombinant (E. coli) and is available in GMP grade for regulated applications. This enzyme is manufactured in an ISO 13485-certified UCF.ME™ facility and meets GMP-grade quality standards. Each lot is subjected to activity assay, purity assessment by SDS-PAGE, and functional validation prior to release. A Certificate of Analysis (CoA) and Safety Data Sheet (SDS) are available on request.

A 5′ cap structure (typically 7-methylguanosine, m7G) on mRNA is essential for ribosomal recognition during cap-dependent translation initiation via eIF4E binding. It also protects mRNA from 5′ exonuclease degradation and signals to the innate immune system that the transcript is "self." Cap-1 structures (2′-O-methylation at the +1 nucleotide) further reduce immunogenicity, which is critical for therapeutic mRNA vaccines and protein replacement therapies.
