Target M-CSF

M-CSF (Macrophage Colony-Stimulating Factor, CSF-1) is a master regulator of macrophage differentiation, polarization, and function. It signals through the M-CSF receptor (M-CSFR / CSF-1R), a receptor tyrosine kinase whose dimerization and autophosphorylation activate NF-κB, JAK/STAT, PI3K/Akt, MAPK/ERK, and IL-10/TGF-β pathways. As an N- and O-glycosylated homodimeric glycoprotein, M-CSF requires correct glycan patterning and disulfide bridging for potent M-CSFR engagement.

Bioactivity Assay: Dose-dependent fold induction of M-NFS-60 cells (ATCC 30-2001) over 20 hr, assessed by Prestoblue™ Cell Viability luminescence. MycoFold rHu M-CSF EC50 <4 ng/mL. Compared head-to-head against Peprotech (E. coli, AF-300-25) and Proteintech (HEK-293, HZ-1192) — MycoFold achieves ~2× higher specific activity than E. coli with equivalent bioactivity to HEK-293 at 3× lower cost per IU.

Thermal Stability: Tm = 79.2°C (Biotium GloMelt™ Kit, Cat. No. 33022-T; triplicate replicates; 30–100°C scan). This exceptionally high melting temperature — 6.6°C above Goat IgG control (72.6°C) — reflects the contribution of glycosylation and disulfide bonds to thermodynamic stability. Reconstituted protein keeps at 2–4°C for up to 4 weeks.

Stability Data: Specific activity measured freshly reconstituted, after 1× freeze-thaw (130% retention), and at 30 days at 4°C (108% retention). Full stability profile documented in lot-specific CoA and comprehensive data packages.

Purity Methods: HP-SEC (Dionex Ultimate 3000; XBridge Premier Protein SEC, 250 Å, 2.5 µm; UV280); microfluidic capillary electrophoresis (mCE, GXII CaliperLS/Perkin Elmer); and SDS-PAGE (Bio-Rad Precision Plus standards). Reconstituted MycoFold rHu M-CSF is stable for up to 4 weeks (28 days) at 2–8°C. Lot-specific stability testing confirmed 108% specific activity retention at 30 days at 4°C (relative to freshly reconstituted protein, set at 100%). For storage beyond 4 weeks, aliquot and store at −20°C. Avoid repeated freeze-thaw cycles — each cycle should be kept to a minimum.
