PDGF-BB (Platelet-Derived Growth Factor BB) is a potent mitogen with pleiotropic roles in mesenchymal and stromal cell biology. It signals through PDGFRα and PDGFRβ receptor tyrosine kinases, activating RAS/MAPK, PI3K/Akt, and PLCγ/PKC cascades to drive cell proliferation, migration, and survival. Uniquely among PDGF isoforms, PDGF-BB binds and activates all three receptor combinations — PDGFRαα, PDGFRαβ, and PDGFRββ — enabling broad targeting across connective tissue, vascular, and progenitor cell populations.

Bioactivity Assay: Dose-dependent stimulation of M-NFS-60 cell proliferation (ATCC Cat. No. 30-2001) over 72 hr was assessed by Prestoblue™ Cell Viability luminescence. MycoFold rHu PDGF-BB achieved an EC50 of 7.2–8.2 ng/mL, with specific activity of ≥1.36×10⁵ IU/mg — over 4.4× higher than the leading E. coli-expressed competitor (3.10×10⁴ IU/mg) and >6.3× greater bioactivity per research dollar.
Thermal Stability: Melting temperature (Tm) was determined in triplicate using the Biotium GloMelt™ Thermal Shift Protein Stability Kit (Cat. No. 33022-T), measuring relative fluorescence by RT-PCR across 30–100°C. MycoFold rHu PDGF-BB Tm = 43°C (vs. Goat IgG control Tm = 72.6°C). Native disulfide bridges and proper 3D folding maintain bioactivity after reconstitution and following refrigerated storage or a single freeze-thaw cycle.
Stability Testing: Natively folded MycoFold rHu PDGF-BB maintains its high bioactivity after 1× freeze-thaw and after refrigerated storage (verified by lot-specific CoA specific activity data).

Unlike E. coli, this eukaryotic system supports chaperone-guided protein folding in the ER and lysis-free secretion, enabling proper disulfide bridge formation and native-like 3D structure without animal-origin inputs or bacterial endotoxin contamination.
In head-to-head testing against the leading E. coli-expressed PDGF-BB, MycoFold achieved >4.4× higher specific activity (1.36×10⁵ vs 3.10×10⁴ IU/mg) and >6.3× greater bioactivity value per research dollar (43.6 vs 7.0 IU/mg/$). Reconstitute lyophilized rHu PDGF-BB in deionized water to 0.1–0.5 mg/mL. No special buffers, carrier proteins, or supplements are required.
