In Vitro
Partially purified porcine kidney N-acetyl-β-D-glucosaminidase is inhibited by Nagstatin potently, specifically and competitively, (IC50 of 0.0012 μg/mL; Ki of 1.7×10-8 M)[1]. Nagstatin inhibits N-acetyl-β-D-glucosaminidase from several sources: Spodoptera litura (IC50 5.85 nM; Ki 1.10 nM), Aspergillus oryzae (IC50 10.6 nM; Ki 9.69 nM), Penicillium oxalicum (IC50 60.6 nM; Ki 8.11 nM), human placenta (IC50 1.83 nM; Ki 0.952 nM), bovine kidney (IC50 1.71 nM; Ki 1.64 nM), and jack bean (IC50 1.87 nM; Ki 1.41 nM)[2].
In Vivo
Nagstatin (250 mg/kg; i.p.; single dose) shows low acute toxicity in mice, with no mortality at a single intraperitoneal dose of 250 mg/kg[1]. In normal mice, Nagstatin potentiates the cellular immune response at a potency comparable to bestatin[1]. In tumor-bearing mice, Nagstatin reactivates a depressed cellular immune response at a potency comparable to bestatin[1].
