Compound Overview
Necrostatin-5, also known as Nec-5, is a potent inhibitor of necroptosis with an EC50 of 0.24 μM. It also inhibits RIP1 and has been reported to show cardioprotective effects[1][2][3]. It is supplied as a white to off-white solid (C19H17N3O2S2, MW 383.49) at 99.40% purity.

In Vitro
| Solvent | Solubility | Notes |
|---|---|---|
| DMSO | 5 mg/mL (13.04 mM) | requires sonication and warming and heat to 60°C; use freshly opened DMSO (absorbed moisture lowers solubility) |
Aliquot the stock solution and store it at -80°C (up to 6 months) or -20°C (up to 1 month); avoid repeated freeze-thaw cycles.
Data provided by the manufacturer.
In Vitro
In a dose-dependent manner, Necrostatin-5 (1, 5, 10, 50, 100 μM) reduces LDH release[3]. At 100 μM, Necrostatin-5 shields MH-S cells from challenge with S. aureus, Listeria monocytogenes, S. pneumoniae, and uropathogenic Escherichia coli (UPEC), and also guards against pneumolysin-induced death[3].
In Vivo
In the isolated rat heart model, Necrostatin-5 (2.46 mg/kg; i.p.) shows cardioprotective effects[2].
| Animal Model | 200-300 g, male Wistar rats[2] |
|---|---|
| Dosage | 2.46 mg/kg |
| Administration | I.p.; 60 min before experiment |
| Result | Reduced the infarction zone caused by 30-min global ischemia and 120-min reperfusion. |
