Neuroligin 4 (NLGN4) is a neuronal cell surface protein that contributes to the formation and remodeling of synapses in the central nervous system. As a member of the neuroligin family, NLGN4 binds to presynaptic neurexins, facilitating the development of functional synaptic junctions.
NLGN4 is implicated in the specification of inhibitory synapses and is enriched in postsynaptic densities. It interacts with scaffolding proteins such as DLG4 (PSD-95), influencing synaptic stability and receptor clustering. Mutations in NLGN4 have been linked to autism spectrum disorders and intellectual disability, highlighting its role in neurodevelopmental processes.
In neurodegenerative disease, synaptic dysfunction is a key early event. Altered NLGN4 expression or function may disrupt inhibitory signaling, contributing to excitotoxicity and network imbalance. Its involvement in synaptic maintenance suggests that NLGN4 may serve as a biomarker for early synaptic pathology and a potential target for therapeutic intervention.
Understanding NLGN4’s role in synaptic regulation offers valuable insights into the molecular basis of cognitive decline and neurodegeneration.
1 µg/ml of SMC-469 was sufficient for detection of Neuroligin 4 in 20 µg of mouse brain lysate by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.
