In Vitro
In RAW 264.7 macrophages, NF-κB-IN-9 (TR2) (1 nM-10 μM) inhibits NO production with an EC50 value of 68.9 nm[1]. In MCF-7 cells, NF-κB-IN-9 (10 μM; 30 s ultrasonic treatment) inhibits proliferation and induces apoptosis. NF-κB-IN-9 is sonocytotoxic to MCF-7 cells, with IC50s of 1.6 μM, 0.55 μM, 0.29 μM at ultrasonic treatment times of 30 s, 2 min, 5 min, respectively[1].
In Vivo
In xenograft mouse models, NF-κB-IN-9 (TR2) given at 1.5 mg/kg (iv; at day 0 and 10, respectively) displays anticancer efficacy together with good tolerance. Fluorescence from NF-κB-IN-9 can be detected in the brain, suggesting that TR2 could penetrate the blood-brain barrier[1].
