Enzyme Activity Assay Kit

Enzyme & Substrate

Nicotinamide N-methyltransferase consumes SAM to methylate nicotinamide, and its overexpression in tumours and adipose tissue redirects methyl-group and NAD+ metabolism.

Assay Principle

The NNMT Activity Fluorometric Assay Kit applies a fluorometric format to this measurement, read at Ex/Em = 400/465 nm.

NNMT (Nicotinamide N-Methyltransferase) is an enzyme that catalyzes the methylation of nicotinamide (NA) and other pyridines using S-adenosyl-L-methionine (SAM) as the methyl group donor to produce SAH and 1-methylnicotinamide (MNA).

Measuring endogenous NNMT activity reports on methyl-group and NAD+ handling in tumour and adipose tissue.

Storage & Handling

Store the Reagent at -20°C (-80°C for enzyme NNMT and SAH Enzyme Mix), protected from light. Shelf life: 6 months.

One or more components require a colder storage temperature than the rest of the kit. Separate them on arrival and file each at the temperature stated above.

Safety

For research use only. Not for use in diagnostic or therapeutic procedures. Handle in accordance with good laboratory practice; consult the SDS before use.

Q.Which reader is required for this activity assay?
A.This kit requires a fluorescence-capable plate reader. The readout is taken at Ex/Em = 400/465 nm.
Q.What does the activity measurement tell me?
A.Measuring endogenous NNMT activity reports on methyl-group and NAD+ handling in tumour and adipose tissue.
Q.Why measure NNMT Activity?
A.Nicotinamide N-methyltransferase consumes SAM to methylate nicotinamide, and its overexpression in tumours and adipose tissue redirects methyl-group and NAD+ metabolism.
Q.Which sample types are validated for this enzyme?
A.The datasheet does not restrict the assay to a named sample list. Prepare lysates or fluids by your standard route and confirm performance with a spike-recovery check on first use.
Q.How should the kit be stored on arrival?
A.Store the Reagent at -20°C (-80°C for enzyme NNMT and SAH Enzyme Mix), protected from light. Shelf life: 6 months. Unpack promptly rather than leaving the box at the bench, and return components to storage immediately after each use.
