Inhibitor Screening Assay Kit

Screening Context

Because NNMT overexpression is linked to obesity and tumour progression, and because its product 1-methylnicotinamide is stable and easily measured, the enzyme has become an attractive small-molecule target.

Assay Principle

The NNMT Inhibitor Screening Colorimetric/Fluorometric Assay Kit applies a dual colorimetric/fluorometric format to this measurement, read at Ex/Em = 400/465 nm.

NNMT (Nicotinamide N-Methyltransferase) is an enzyme that catalyzes the methylation of nicotinamide and other pyridines using S-adenosyl-L-methionine (SAM) as the methyl group donor to produce SAH and 1-methylnicotinamide.

The screening format ranks candidate NNMT inhibitors rather than profiling endogenous enzyme levels.

Storage & Handling

Store the Reagent at -20°C protected from light. Shelf life: 12 months. For Enzyme I and Enzyme II, long term storage at -80°C.

One or more components require a colder storage temperature than the rest of the kit. Separate them on arrival and file each at the temperature stated above.

Safety

For research use only. Not for use in diagnostic or therapeutic procedures. Handle in accordance with good laboratory practice; consult the SDS before use.

Q.What instrument does the screen require?
A.This kit requires either an absorbance or a fluorescence plate reader. The readout is taken at Ex/Em = 400/465 nm.
Q.What does this screening assay tell me?
A.The screening format ranks candidate NNMT inhibitors rather than profiling endogenous enzyme levels.
Q.Why measure NNMT Inhibitor Screening/?
A.Because NNMT overexpression is linked to obesity and tumour progression, and because its product 1-methylnicotinamide is stable and easily measured, the enzyme has become an attractive small-molecule target.
Q.What can I screen against?
A.The datasheet does not restrict the assay to a named sample list. Prepare lysates or fluids by your standard route and confirm performance with a spike-recovery check on first use.
Q.How should the kit be stored on arrival?
A.Store the Reagent at -20°C protected from light. Shelf life: 12 months. For Enzyme I and Enzyme II, long term storage at -80°C. Unpack promptly rather than leaving the box at the bench, and return components to storage immediately after each use.
