Screening Context
Because NNMT overexpression is linked to obesity and tumour progression, and because its product 1-methylnicotinamide is stable and easily measured, the enzyme has become an attractive small-molecule target.
Assay Principle
The NNMT Inhibitor Screening Colorimetric/Fluorometric Assay Kit applies a dual colorimetric/fluorometric format to this measurement, read at Ex/Em = 400/465 nm.
NNMT (Nicotinamide N-Methyltransferase) is an enzyme that catalyzes the methylation of nicotinamide and other pyridines using S-adenosyl-L-methionine (SAM) as the methyl group donor to produce SAH and 1-methylnicotinamide.
The screening format ranks candidate NNMT inhibitors rather than profiling endogenous enzyme levels.
