Compound Overview

NTZ-24 is a selective inhibitor of the STAT3 pathway. It suppresses STAT3 phosphorylation at Tyr705, blocks the STAT3-DNA interaction, and downregulates STAT3 downstream target proteins, while also inducing cell-cycle arrest and promoting apoptosis in cancer cells. Against HeLa cells it exerts significant antiproliferative activity (IC50 = 3.3 μM), and can be used in cervical cancer research[1]. It has a molecular formula of C10H5Cl2N3O3S and a molecular weight of 318.14 g/mol.

In Vitro

In HEK-Blue IL-6 cells, NTZ-24 inhibits the IL-6/JAK/STAT3 pathway with an IC50 of 0.8 μM[1]. NTZ-24 (48 h) suppresses the proliferation of HeLa epithelial carcinoma cells, with an IC50 of 3.3 μM[1]. In human normal embryonic kidney HEK 293T cells, NTZ-24 (10-50 μM; 48 h) shows low cytotoxicity, with cell viability inhibition of 8.0% at 10 μM and 19.1% at 50 μM[1].
