Product Overview
Nucleic acids interfere with many steps of protein work, including cell lysis, chromatographic purification, protein quantification, activity assays and crystallization, especially when the protein of interest binds DNA or RNA. The Nucleic Acid Removal Kit removes nucleic acids from a protein solution or cell lysate to below 250 pg/ml.
NAR Reagent 1 and Reagent 2 are ionic polymers that precipitate nucleic acids, which are then removed by centrifugation. The final solution contains 0.45 M NaCl, and the reagents are easily removed during subsequent protein purification.

Protocol Outline
- Keep all solutions at 0–4°C. Mix one volume of NAR Reagent 1 with 9 volumes of cell suspension (then lyse) or protein solution (then incubate on ice for 10 min).
- Remove insoluble material or precipitate by centrifugation at 6000 x g for 20 min at 4°C.
- Add one volume of NAR Reagent 2, incubate on ice for 5 min and remove the precipitate by centrifugation at 6000 x g for 20 min at 4°C.
- Continue with protein purification; for ion exchange chromatography, dialyze against a low-salt buffer first.
