ODN 1826 is a class B CpG oligodeoxynucleotide (ODN) that acts as a TLR9 agonist. It induces NO and iNOS production and enhances apoptosis, boosts immune surveillance, and increases aortic atherosclerotic plaque size; it has also shown antitumor activity against lung cancer, glioma, and melanoma[1][2][3][4][5][6]. In Vitro

In RAW 264.7 mouse macrophages, ODN 1826 (1 μg/mL; 24 h) induces production of nitric oxide (NO) and inducible nitric oxide synthase (iNOS)[3].

In Vivo

Intratumoral injection of CpG-1826 (100 μg/tumor) every 3 days for up to 4 times slows the growth of subcutaneously implanted GL261 gliomas in C57BL/6NTac mice, with increased infiltration of macrophages and B cells into the tumors and significant growth delay in ~50% of mice[4]. In a chronic vascular injury mouse model, 18 nM ODN 1826 given s.c. 3 times a week for 7 weeks enlarges aortic atherosclerotic plaques[5]. In a Lewis lung cancer mouse tumor model, ODN 1826 (0.05 mg i.p. on days 1, 3, 5, 8, 11, 13) exerts a strong anti-tumor growth effect[6].

Achieved complete tumor regression and induced long-term immune memory in combination with Cyclophosphamide. Delayed growth but did not induce regression, with modest increases in F4/80+ macrophages and B220+ B cells in combination with Cyclophosphamide (in B16F10 melanoma model).
