In Vitro
ODN 2088 (0.01-10 μM; 48 h) is not cytotoxic to human PBMCs[1]. ODN 2088 (0.01-10 μM; 24 h) inhibits IFN-α release from human PBMCs stimulated with CpG-ODN 2216 (3 μM) and the TLR7-ligand RNA-ORN 22075 (5 μM)[1]. In human PBMCs, ODN 2088 (0.01-10 μM; 48 h) barely inhibits IL-6 release stimulated with CpG-ODN 2006 (100 nM) but does inhibit IL-6 release stimulated with Imiquimod (5 μg/ml)[1]. ODN 2088 (0.1-10 μM; 24 h) barely inhibits IL-6 release from B-cells stimulated with CpG-DNA 2006 (100 nM) but inhibits IL-6 release from human B-cells stimulated with Imiquimod (5 μg/ml)[1]. ODN 2088 presumably impairs the TLR9-induced signaling triggered by CpG-ODNs through competitive binding to the C-terminal fragment of TLR9[1]. In BMDMs stimulated with CpG-ODN 1826 (100 nM), ODN 2088 (0.001-10 μM; 24 h) inhibits TNF-α secretion in a dose-dependent manner, and it shows only weak inhibition when the cells are stimulated with the TLR7-agonist imiquimod[3]. ODN 2088 (10 μM) stimulates B cell proliferation[3].
