Compound Overview
Oleocanthal is an orally active phenolic seciridoid compound that can be extracted from olive oil. It inhibits COX-1 and COX-2, reduces ROS and NO, and upregulates Nrf-2 and HO-1, and it also reduces Aβ deposition. It shows anti-Leishmania activity against promastigotes and amastigotes of L. major, with IC50 values of 18.7 and 87 μg/mL, respectively, and exhibits anticancer activity against colon, breast, liver, and melanoma cancers as well as anti-inflammatory and neuroprotective properties; it can be used in Alzheimer's disease research[1][2][3][4][5][6]. It is supplied as a white to off-white solid (C17H20O5, MW 304.34) at 95.04% purity and is derived from the plant Canarium album (Lour.) Rauesch.

In Vitro
In LPS-stimulated murine peritoneal macrophages, Oleocanthal (25-100 μM) lowers levels of ROS, nitrites, and pro-inflammatory cytokines[1]. Oleocanthal (1-25 μM; 12 h) reduces LPS-induced NOS2 synthesis and NO production in ATDC-5 chondrocytes[2]. Growth of L. major promastigotes and amastigotes is inhibited by Oleocanthal (0-200 μg/mL; 48-60 h), with IC50 values of 18.7 and 87 μg/mL, respectively[5]. Inhibitory activity against COX-1 and COX-2 enzymes is shown by Oleocanthal[6].

In Vivo
5xFAD mice given Oleocanthal (10 mg/kg/day; p.o.; daily; 3 months) show better metabolic and behavioral phenotypes, lower Aβ deposition, and improved BBB function[3]. In BALB/c mice infected with L. major, Oleocanthal (5 mg/kg b.w., i.p., 16 total doses given 3 times/week) lowers footpad thickness and parasite load and induces Th1-type immunity[5].
