Enzyme

Bacterial DNA topoisomerase II (gyrase) is a type II topoisomerase that converts relaxed circular DNA into negatively supercoiled DNA. Type II topoisomerases cut and rejoin both DNA strands to change DNA topology.

Some inhibitors, such as fluoroquinolones, block the rejoining step; the enzyme then stays covalently linked to cut DNA in a cleavage complex. Ciprofloxacin and novobiocin both inhibit gyrase, but only ciprofloxacin produces cleavage complexes.

This product is the GyrA subunit of P. aeruginosa DNA gyrase. DNA gyrase is active as a heterotetramer of two subunit types, so GyrA alone has no supercoiling activity; combine it with GyrB (P. aeruginosa GyrB, BHP16300034) to reconstitute the active enzyme. The DNA gyrase holoenzyme is also sold separately as P. aeruginosa DNA Gyrase, BHP16300032.

Safety & Handling

For Research Use Only (RUO). Not for use in diagnostic or therapeutic procedures. This enzyme is shipped frozen. Follow standard laboratory safety practices.
Q.What is supplied?
A.50 µl of 2 µM P. aeruginosa GyrA (catalog # GYRA-100PA), enough for 100 assays.
Q.Can this subunit be used on its own?
A.No. DNA gyrase needs both subunit types for its DNA reactions. Combine GyrA with GyrB (BHP16300034), or use P. aeruginosa DNA Gyrase (BHP16300032), which is the enzyme used in the matching ProFoldin assay kits.
Q.Is a datasheet or protocol available?
A.There is no separate datasheet for this product. Please contact us for recommended reaction conditions.
Q.How is it shipped and stored?
A.This enzyme is shipped frozen. Please contact us to confirm long-term storage conditions.
