Toward a peptide substrate, the catalytic activity of PDK1 is stimulated sixfold by PDK1-IN-RS2. A salt bridge links the sulfonyl group of PDK1-IN-RS2 to Arg131, since the sulfonamide is likely ionized under the crystallization conditions[1].
