Compound Overview
Pentixafor is a cyclic pentapeptide ligand that targets CXCR4. It comprises the cyclic pentapeptide cyclo(dTyr-dOrn(NMe)-R-Nal-G), a linker, and a DOTA chelator, and can be labeled with 68Gallium (68Ga) for positron emission tomography (PET) imaging[1][2][3][4]. It can be used for the synthesis and research of Radionuclide-Drug Conjugates (RDCs). It is supplied as a white to off-white solid (C60H80N14O14, MW 1221.36) at 99.60% purity.

In Vitro
| Solvent | Solubility | Notes |
|---|---|---|
| Methanol | ≥ 125 mg/mL (102.34 mM) | — |
| DMSO | 100 mg/mL (81.88 mM) | requires sonication; use freshly opened DMSO (absorbed moisture lowers solubility) |
Aliquot the stock solution and store it at -80°C (up to 6 months) or -20°C (up to 1 month); sealed storage, away from moisture and light; avoid repeated freeze-thaw cycles.

In Vitro
In CXCR4-transfected Chem-1 cells, [18F]AlF-NOTA-Pentixather (195 pmol, 60 minutes) shows significant internalization and high cellular uptake[4].
In Vivo
In the PC-3 prostate cancer xenograft model, [68Ga]Pentixafor (15 MBq, single intravenous dose) is suitable for PET imaging[2]. In the Daudi lymphoma xenograft SCID mouse model, [18F]AlF-NOTA-Pentixather (0.30 MBq, single intravenous dose) can specifically target the CXCR4 receptor for PET imaging[4].
