Compound Overview
Pentosan Polysulfate Sodium (W/W 43%) is an orally bioavailable, semi-synthetic medication with anti-inflammatory and pro-chondrogenic properties. It is also a potent, selective anti-HIV agent and is used for the treatment of interstitial cystitis[1][2][3]. It is supplied as a white to off-white solid (MW 4000-6000) at 43% purity.

Safety
For Research Use Only. Not for use in diagnostic or therapeutic procedures, and not for human or veterinary use. Handle in accordance with the Safety Data Sheet and your institution's chemical hygiene plan.
In Vitro
Pentosan Polysulfate Sodium has been shown to inhibit HIV-1 activity in MT-4 cells, with an ED50 of 0.19 μg/mL. In HUT-78 cells, it inhibits HIV-1 antigen expression with an ED50 of 0.02 μg/mL, and HIV-1 antigen expression is fully inhibited at a concentration of 4.0 μg/mL[2]. Pentosan Polysulfate Sodium suppresses NF-κB, lessens the proinflammatory actions of TNFα, and reduces MCP-1 production stimulated by high glucose and advanced glycation end products (AGEs)[3].
In Vivo
Pentosan Polysulfate Sodium has been shown to reduce interstitial inflammation and glomerulosclerosis in 5/6 nephrectomized rats. Treatment with pentosan polysulfate preserves renal function, significantly lowers albuminuria, and markedly lessens the severity of renal lesions, including tubulointerstitial inflammation. In aging diabetic kidneys, Pentosan Polysulfate Sodium additionally curbs the upregulation of TNFα and proinflammatory genes[3].
