Scientific Background

ITCH (itchy E3 ubiquitin protein ligase) encodes a member of the Nedd4 family of HECT domain E3 ubiquitin ligases. HECT domain E3 ubiquitin ligases transfer ubiquitin from E2 ubiquitin-conjugating enzymes to protein substrates, thus targeting specific proteins for lysosomal degradation. The encoded protein plays a role in multiple cellular processes including erythroid and lymphoid cell differentiation and the regulation of immune responses. Mutations in ITCH are a cause of syndromic multisystem autoimmune disease. Alternatively spliced transcript variants encoding multiple isoforms have been observed for ITCH.

Alternative Names

ITCH; E3 ubiquitin-protein ligase Itchy homolog; Itch; Atrophin-1-interacting protein 4; AIP4; NFE2-associated polypeptide 1; NAPP1

What species does this antibody react with?
This antibody has been validated for reactivity against Human, Mouse. Actual cross-reactivity is determined by immunogen sequence homology across species. Contact technical support for unlisted species.
What applications has this antibody been validated for?
Validated applications include: Immunohistochemistry (IHC), ELISA. Each application was tested with positive and negative controls. Contact our team regarding applications not listed.
How was this antibody produced and purified?
This polyclonal IgG antibody was raised in rabbit against a synthesized target-specific peptide. It was affinity-purified from antiserum using an epitope-specific immunogen, yielding a highly specific antibody suitable for multiple research applications.
What is the expected molecular weight of ITCH?
The predicted molecular weight of ITCH is ~102.8 kDa. Band size may vary slightly depending on post-translational modifications and gel percentage.
What storage conditions does this antibody require?
Store at −20°C for long-term storage. Avoid repeated freeze-thaw cycles. Working aliquots can be stored at 4°C for short-term use. The formulation includes 50% glycerol as a cryoprotectant.
Is this antibody specific for the phosphorylated form of ITCH?
Yes. This antibody is designed to detect ITCH only at the phosphorylation site specified in the product name. Lambda phosphatase treatment eliminates the signal, confirming phospho-specificity.
