Scientific Background

BRCA1 (BRCA1, DNA repair associated) encodes a nuclear phosphoprotein that plays a role in maintaining genomic stability, and it also acts as a tumor suppressor. The encoded protein combines with other tumor suppressors, DNA damage sensors, and signal transducers to form a large multi-subunit protein complex known as the BRCA1-associated genome surveillance complex (BASC). This gene product associates with RNA polymerase II, and through the C-terminal domain, also interacts with histone deacetylase complexes. This protein thus plays a role in transcription, DNA repair of double-stranded breaks, and recombination. Mutations in BRCA1 are responsible for approximately 40% of inherited breast cancers and more than 80% of inherited breast and ovarian cancers. Alternative splicing plays a role in modulating the subcellular localization and physiological function of this gene. Many alternatively spliced transcript variants, some of which are disease-associated mutations, have been described for BRCA1, but the full-length natures of only some of these variants has been described. A related pseudogene, which is also located on chromosome 17, has been identified.

What species does this antibody react with?
This antibody has been validated for reactivity against Human, Rat. Actual cross-reactivity is determined by immunogen sequence homology across species. Contact technical support for unlisted species.
What applications has this antibody been validated for?
Validated applications include: Western Blot (WB), Immunohistochemistry (IHC), ELISA. Each application was tested with positive and negative controls. Contact our team regarding applications not listed.
How was this antibody produced and purified?
This polyclonal IgG antibody was raised in rabbit against a synthesized target-specific peptide. It was affinity-purified from antiserum using an epitope-specific immunogen, yielding a highly specific antibody suitable for multiple research applications.
What is the expected molecular weight of BRCA1?
The predicted molecular weight of BRCA1 is ~207.7 kDa. Band size may vary slightly depending on post-translational modifications and gel percentage.
What storage conditions does this antibody require?
Store at −20°C for long-term storage. Avoid repeated freeze-thaw cycles. Working aliquots can be stored at 4°C for short-term use. The formulation includes 50% glycerol as a cryoprotectant.
Is this antibody specific for the phosphorylated form of BRCA1?
Yes. This antibody is designed to detect BRCA1 only at the phosphorylation site specified in the product name. Lambda phosphatase treatment eliminates the signal, confirming phospho-specificity.
