Product Overview
The Plant/Fungal DNA Extraction Kit isolates and purifies total DNA from 100–500 mg of fresh plant tissue, 20–50 mg of dried plant tissue or 100–300 mg of fungi. The DNA is eluted in 100–200 µL of preheated Buffer TE. The expected purity is about OD260/OD280 1.8–2.0.
Samples are lysed at 65°C for 30 min, extracted with chloroform at 12,000 rpm for 5 min and precipitated with isopropanol. The mixture is bound to adsorption column P at 12,000 rpm for 30 s, washed once with Buffer RPB and twice with Wash Buffer WB, dried at 14,000 rpm for 1 min and eluted with preheated Buffer TE after standing 2 min at room temperature.

Storage & Handling
Buffers PGE, RPB, WB and TE and the adsorption columns are stored at room temperature for 1 year. RNase A is stored at -20°C for 1 year.
Usage Notes
- Avoid repeated freeze-thaw cycles of the sample.
- Lyse the sample fully to prevent the column from blocking.
- For low yield, increase the sample amount proportionally within the stated range.
- If precipitation occurs in a buffer, pre-incubate it at 37°C before use.
- Confirm that ethanol has been added to the wash buffers before use.
- Allow 2–5 minutes for the DNA to dissolve during elution for best results.
- Chloroform and isopropanol are required but are not supplied with the kit.
