Product Overview

This kit uses a unique buffer system and is especially suitable for extracting genomic DNA from fresh plant materials. It can remove impurities, proteins and other organic compounds in cells to the greatest extent. The kit yields high-molecular-weight genomic DNA of high purity. The DNA recovered by this kit can be used in various routine operations, including enzyme digestion, PCR, library construction, Southern hybridization and so on.

Plant tissue of 100 mg is ground under liquid nitrogen and lysed in 600 µL of Buffer PGE for 20 min at 60°C. The lysate is extracted with 700 µL of chloroform and centrifuged at 12,000 rpm for 10 min; DNA in the aqueous phase is precipitated with isopropanol, washed twice with 75% ethanol, air-dried and dissolved in 50–200 µL of Elution Buffer. Purified DNA has an OD260/OD280 of 1.7–1.9.

Usage Notes

  • Avoid repeated freezing and thawing to prevent DNA fragmentation.
  • Buffer PGE may turn yellow, which does not affect the extraction result.
  • Chloroform, isopropanol and 75% ethanol are used in the protocol but are not supplied in the kit; handle them in a fume hood.
  • Air-dry the residual ethanol before dissolving the DNA.
  • A larger dissolution volume increases yield but lowers the DNA concentration.
